Diet and K-ras mutations in colorectal cancer

A hospital-based case-control study was conducted during 1996-98 to examine the relationship between diet and K-ras mutations in colorectal cancer [Spain]. The diet of 424 cases and 401 controls were assessed using a structured dietary history questionnaire. Tumour samples from 419 cases were screened for mutations in K-ras oncogene by polymerase chain reaction and single-strand conformation polymorphism separation. Mutations in K-ras were observed in 39% of cases. For all cases, protective factors were consumption of fruits and vegetables. The protective effect of high fruit and vegetable consumption was stronger in cases with wild type K-ras than in cases with mutated K-ras. Risk was increased with pastry, alcohol and beef intake. No differences according to K-ras status were observed for the increase in risk for alcohol or beef intake. However, for pastry, the increase in risk was higher in cases with mutated K-ras than in cases with wild type K-ras. It is concluded that there is interaction between diet and K-ras mutations in colorectal cancer.
