Influence of vitamin A acetate, alcohol and their radiolytic products on the in vitro labilization of lysosomes and tissue autolysis

Lamb liver homogenate, 10% w/v, remained unirradiated or received gamma -radiation in doses of 0.1, 0.25 or 0.5 Mrad, and was allowed to autolyse for up to 3 h at 37 deg C. The release of acid ribonuclease, cathepsin and aryl sulphatase was progressively enhanced by increasing doses of radiation; acid ribonuclease showed the greatest free activity and aryl sulphatase the least in all samples including controls. Hydrolytic end products including peptides and amino acids, inorganic phosphate and 260E absorbing compounds also increased with increasing radiation dose. Retinyl acetate was irradiated and eluted to obtain degradation products and fraction F so obtained enhanced the release of acid ribonuclease and cathepsin free activities from homogenate, both initially and during autolysis, whereas retinyl acetate, irradiated or not, had no significant effect. Ethanol, 20 mu g/ml, retinyl acetate and retinol had no effect on enzyme release from the lysosome-rich fraction of lamb liver during incubation, but fraction F induced the release of both free and bound activities of acid ribonuclease and aryl sulphatase, the former to a greater extent.
