Synthesis of chimeric IgE in vivo after ballistic transfection of immunoglobulin genes in various mouse tissues

Genes of heavy and light chains of chimerical (mouse/human0 immunoglobulin E were inserted into various tissues of mice by means of ballistic transfection in vivo. The concentration of full-size chimerical IgE in blood serum was assesed by immunoassay. It was shown that after the transfection of IgE gene in to cartilage tissue, blood IgE concentration was several times lower than after the transfection of liver, spleen and paw cushions. At the same time, the content of antibodies to IgE xenogenic determinants did not depend on tissue type. After repeated transfection on the 114th day, neither chimerical IgE nor antibodies to it were detected. The results of the study can be used to select conditions for genetic immunization.
