Antifertility effect of three new clomiphene analogues on animals

3 Clomiphene analogues were administered orally by gavage to proven male and female swiss albino mice, wistar rats and colony bred rabbits on different days postcoitum. Vaginal smears were performed on all animals. Laparotomy to observe the changes or the presence of implantation sites was performed on all animals. EIPW III at a single dose of 3 and 4 mg/kg had a 100% antifertility effect when given pre-and postimplantation. The drug did not effect male fertility. The estrogenicity of the drug was indicated by the change in vaginal cytology, i.e. typical estrus smears 48 hours after drug treatment during pregnancy; cornified cells after 24 hours and highly vascularized fluid-filled uteri on laparotomy. Clomiphene citrate, at a single oral dose of 3 mg/kg preimplantation in mice had 100% antifertility effect. EIPW 113 and 103 had no appreciable antifertility effect at the same dose.
