Curcumin, an active principle isolated from Curcuma longa, possesses significant anti-inflammatory activity. In a dose of 100 mg/kg (2 .times. anti-inflammatory ED50) administered orally for 6 consecutive days, curcumin produced gastric ulceration in albino rats. Marked reduction in the mucin content of gastric juice seems to be responsible for this ulcerogenic response. Pretreatment with adrenergic, cholinergic, tryptaminergic and histaminergic (H1) receptor antagonists provided partial protection against curcumin-induced gastric ulcers; metiamide pretreatment, however, almost completely prevented the development of gastric lesions and prevented decrease in the mucin secretion. H2 receptors are apparently predominantly involved in the ulcerogenicity of curcumin. The change in the mucin content appears to be the determinant factor in the ulcerogenicity of curcumin.
