The efficacy of long-term treatment with ranitidine 150 mg at bedtime in maintaining the healing of duodenal ulcer was tested in 22 patients after successful short-term therapy with ranitidine at the full dose of 300 mg/day. The trial was completed by 16 subjects (6 dropouts). The cumulative recurrence rate during 1 yr of treatment was 25%; remission persisted in 75% of the patients. No statistically significant differences were noted in mean fasting serum pepsinogen and trypsinogen values during and after treatment as compared to mean pretreatment values. There was a statistically significant rise in mean fasting serum gastrin level from 156 .+-. 27 SD to 258 .+-. 110 SD pg/ml (P < 0.01) only during the 12th mo. of therapy. This increase promptly subsided to normal levels after cessation of therapy (162 .+-. 34 SD pg/ml). No appreciable side effects were noted.
