Generation of homozygous transgenic lines facilitates breeding control and subsequent analysis in gene transfer experiments. Identification of homozygous animals by a semiquantitative PCR technique is described. This uses ethidium bromide stain and visual evaluation of amplification products. Optimization was carried out to distinguish between homozygous and hemizygous mice transgenic for wild-type murine tyrosinase gene construct ptrTyr4. The procedure can also be used for breeding control and, in a modified form, for classifying different mouse lines in regard to transgene copy number.
