In comparison with muscle from normal pigs, muscle slices from a strain of pigs susceptible to malignant hyperthermia showed greater depletion of ATP and greater production of lactate; halothane increased the ATP depletion and lactate production in vitro. No difference between the calcium uptake capacities of sarcoplasmic reticulum from normal and the susceptible pigs was detected. Halothane increased the amounts of Ca++ bound by sarcoplasmic reticulum from both normal and susceptible muscle. It is suggested there is an underlying myopathy in susceptible pigs, the nature of which appears to be a rapid utilization of muscle ATP and loss of glycolytic control mechanisms.
