Induction of tumors and malformations in mice after prenatal treatment with some antibiotic drugs

Four antibiotic drugs, 5-nitro-2-furaldehyde semicarbazone (nitrofurazone), N-(5-nitro-2-furfurylidene)-1-aminohydantoin (nitrofurantoin), 7-(thiopene-2-acetamido) cephalosporanate (cephalothin) and streptomycin sulfate (streptomycin), were tested for teratogenicity in mice. Among them, nitrofurazone, nitrofurantoin and cephalothin induced low but significant yields of malformations, when high doses were given at critical stages (days 9, 10 and 11 of pregnancy). Nitrofurantoin induced low but significant yields of lung tumors in the offspring, when given to pregnant mice on days 13, 15 and 17. Nitrofurazone was not carcinogenic when given transplacentally, but induced significant yields of lung tumors, when given during neonatal and young age.
