Comparative studies of changes in the EEG and behavior over a 24 hour period after the administration of psycho-active drugs to normal volunteers were performed. The effects of four drugs chlorpromazine, thioridazine, clozapine and the indenopyridine YG 19-256 were investigated and compared with their pre-drug control levels and with placebo. Two quantitative computer assisted techniques were employed: power spectral analysis and iterative interval analysis. Results indicated that the three neuroleptics had qualitatively similar effects on the EEG. Nonetheless important differences were also apparent; clozapine in contrast to chlorpromazine and thioridazine, had specific effects on the lower ß band (12-16 cps) an increase in relative power. It was suggested that this phisiological specificity might be related to the unique clinical effects of clozapine strong anti-psycotic action in the absence of the extra-pyramidal side effects which are typical of other neuroleptic drugs. The indenopyridine YG 19-256, was different from the three neuroleptics tested both in its behavioural effects and in its effects on the EEG. From these results it was apparent that the methods employed were capable both of detecting the central actions of these drugs and of discriminating between those with different clinical effects.
