Work in progress on assembling a complete physical map of the genome is reported, the first stage of which involved analysis of ~20 000 random cosmid clones. Using computer matching programs, these clones were assembled into some 750 overlapping groups, encompassing 90-95% of the genome. Attempts are being made to bridge the gaps by selecting missing clones by hybridization. As a complement to the project, a restriction fragment length polymorphism (RFLP) map was constructed which currently contains 175 markers and provides contact between the physical map and classical genetic map. The main aim is to simplify cloning of genes when the locus but not the gene product is known.
