Experimental infections of a Phlebotomus perniciosus colony using different procedures

Sandflies from 2 colonies of P. perniciosus established in Italy and Spain, respectively, were infected with a Leishmania infantum isolate from Spain using a membrane feeding technique. Only the Spanish colony of P. perniciosus was completely susceptible to infection (55/68 of the Italian flies became infected). Experimental infections of P. perniciosus with the same L. infantum isolate were carried out using RPMI or PBS as infective feeding media, and by providing the sandflies with an opportunity to re-feed on an anaesthetized hamster 5-6 days after the initial infection. The results demonstrated that using PBS it was possible to re-feed a large number of Leishmania-infected flies.
