Combination therapy with 5-azacytidine plus beta-2'-deoxythioguanosine in adult acute leukemia

In some laboratory studies, drug sequences are important. In L1210 mouse leukemia, 5-azacytidine followed by .beta.-2'-deoxythioguanosine was no better than the 2 drugs given concurrently or .beta.-2'-deoxythioguanosine followed by 5-azacytidine. Clinically, a limited trial of 5-azacytidine followed by .beta.-2'-deoxythioguanosine showed no practical advantage. Concurrent 5-azacytidine (150 mg/m2 per day) and .beta.-2'-deoxythioguanosine (300 mg/m2 per dose) is tolerable and active in human acute leukemia, but its possible advantage over 5-azacytidine alone requires further testing.
