Rapid identification of recombinant baculoviruses using PCR

The technique of using the polymerase chain reaction to check for isolates in which the polyhedrin gene of the baculovirus has been replaced by a foreign gene of interest, via homologous recombination, is described. Two primers with sequences for the polyhedrin gene are used, plus a primer derived from sequences 460 bp downstream of the start codon of the foreign gene. By using these 3 primers to amplify both wild-type virus DNA and recombinant DNA, the isolates can be checked both for the presence of recombinant virus and for purity of the isolate in a single step.
