Experimental study of the effect of lithium, haloperidol, caffeine and theophylline on convulsive seizure reactions

The effect of different doses of lithium, haloperidol, caffeine and theophylline on convulsive seizure reactions is studied in experiments with albino mice and rats using electric. pentylenetetrazol (Cor.) and strychnine (Str.) convulsion models. The substances have been administered intraperitoneally (i. p.) and intracerebroventricularly (i. c. v.) at different time intervals before the convulsive agent. After intravenous administration in rabbits a study was made of their effect on the spontaneous bioelectrical cortical activity and on the Corazol convulsion threshold. None of the psychoactive drugs tested had a considerable effect on convulsive reactivity during electric and strychnine convulsions. Lithium and haloperidol had a slight inhibitory effect when pentylenetetrazol convulsion models were used. Conversely, caffeine and theophylline potentiated slightly the convulsion effect of the pentylenetetrazol. These effects of the psychoactive substances depend to a certain extent on the dose, the way of administration and the interval between the administration of the psychoactive and convulsive agents.
