The present paper reports the results of a study undertaken to assess the effects of parathyroid hormone (PTH) on renin release in the rabbit. In vivo, injection of an acute dose of PTH produced hypotension and increased immediate and transient plasma renin activity. This increased activity may be ascribable to the hypotensive effects of the hormone or its direct action on renin-secreting renal juxtaglomerular cells. In vitro, PTH stimulated renin release by isolated glomeruli of the rabbit renal cortex, an effect modulated by the amount of extramedular calcium present. The foregoing observations are in agreement with the mechanisms controlling renin release by vasoactive agents reported in the literature. Furthermore, PTH may also play a role in hypertension frequently associated with primary hyperparahyroidism and hyperthyroidism secondary to advanced renal failure.
