Enhancement of insecticide toxicity by the antidiuretic agent, diazoxide

Sheep and pigs were dosed with the antidiuretic compound, diazoxide, and with diazoxide in combination with representative compounds from the major classes of organic insecticides: carbaryl, benzene hexachloride (BHC) and dimethoate. Minimal toxic doses of the insecticides in sheep were unchanged, but diazoxide reduced the minimal toxic doses in pigs; that of carbaryl was lowered from 1, 000 to 50 mg/kg. The toxicity of BHC was enhanced by the administration of diazoxide, the minimal toxic dose being reduced from 50 mg/kg of the gamma isomer to 15 mg/kg. The toxicity of dimethoate was slightly increased. Administration of diazoxide alone in three doses, with 3-week intervals between doses, elicited signs of uraemic poisoning in pigs. Histological examination of renal tissue sections showed dilation of the collecting tubules and cloudy swelling of cells of the proximal convoluted tubules.
