Measurement of blood levels of anesthetic agents by gas chromatography (IV) --direct analysis of the concentration of sevoflurane dissolved in blood

In the present study, we modified our direct method (for gas chromatographic measurement of isoflurane in the whole blood) to measure the concentrations of sevoflurane dissolved in blood. As the optional conditions for gas chromatography, the precolumn was packed with Sunpak A that had a 0.3% coating of silicone GE SE-30 and the main column was packed with Chromosorb W that had a 10% coating of silicone DC-550. With this method, 5 .mu.l of blood containing sevoflurane was injected directly into the BSV using a microsyringe. The sevoflurane and water were completely vaporized, then led into the precolumn to separate the water from the sevoflurane. Next, the water was removed from the precolumn. The sevoflurane was then led into the main column and the concentrations of sevoflurane were measured. The time needed for one test was less than 10 min and more than 100 samples could be measured consecutively. The coefficient of variation for the data was less than 2% and accurate reproducible quantitative measurement of sevoflurane dissolved in blood was achieved. The linearity showing a straight line passing through the 0 point was obtained by this method until the sevoflurane concentration reached about 64 mg/dl.
