Comparison of pharmacological activity of nicotine and related alkaloids occurring in cigarette smoke

The actions of nicotine were compared with the actions of a number of other alkaloids known or suspected to occur in cigarette smoke. The pharmacological tests used were contraction of the isolated guinea pig ileum and frog rectus, rise in blood pressure in the pithed rat, release of catecholamines from the cat adrenal, neuromuscular blockade in the isolated phrenic nerve-diaphragm of the guinea pig and blockade of spinal reflexes in the cat and chick. Nicotine was the most potent of the alkaloids tested in all these actions. The relevance of these observations to the pharmacological effects of smoking is discussed.
