Radioimmunoassay of mitochondrial aspartate aminotransferase in rat tissues

We performed a preliminary radioimmunoassay (RIA) for mitochondrial aspartate: 2-oxoglutarate aminotransferase (EC 2.6.1.1: m-AST) in order to determine the amount of m-AST protein in tissues and sera. Mitochondrial AST was purified from skeletal muscle of Wistar rats. Antiserum for m-AST was prepared in New Zealand white rabbits. The purified enzyme was radioiodinated by the lactoperoxidase method. By this newly developed RIA, m-AST could be measured in the range of 3-200 ng per tube and c-AST exhibited no antibody-dependent binding up to 200 ng per tube. Reliability of the method was confirmed by the observation that the coefficient of variation of simultaneous multiple determinations of m-AST in liver homogenate and serum was from 3.1% to 18%. These results suggest that this method is easily applicable to the quantitative determination of m-AST protein in rat tissues and serum.
