From a study of various rapid techniques for fixing and embedding specimens for electron microscopy (EM), it was concluded that the optimum protocol was 15 minutes fixation in 3% glutaraldehyde, 30 minutes in 2% osmium tetroxide, progressive washings in ethanol and then in propylene oxide, before embedding. Methods are also described for obtaining EM preparations from formalin-fixed, autolysed PM specimens, or from stained frozen or paraffin sections (to correlate EM and optical microscope findings). Examples are given of the use of these techniques.
