Stereoselective determination of racemic pindolol in rat plasma and its application

Plasma concentrations of racemic pindolol, after intravenous injection (5 mg/kg) to 24 hr-fasted rats, were stereoselectively determined on HPLC by diastereometric derivatization with 2,3,4,6-tetra-O-.beta.-D-glucopyranosyl isothiocyanate. The biological half life and apparent volume of distribution for R(+)- and S(-)-pindolol were 19.3 min, 3.39 l/kg and 24.5 min 8.33 l/kg, respectively. The apparent volumes of distribution for R(+)- and S(-)-pindolol were decreased to approximately half of the control by pretreatment with bacterial lipopolysaccharide (LPS) (10 .mu.g/kg i.v.) 24 hr before racemic pindolol injection, but in both instances the biological half lives were changed only slightly. From the dose-dependent curve, the half effective doses of LPS for R(+)- and S(-)-pindolol were found to be 0.01 and 0.1 .mu.g/kg i.v., respectively.
