Ketoconazole (100 mg/kg daily) increased the immunosuppressive activity and toxicity of cyclosporine (25-200 mg/kg daily) in mice. Ketoconazole alone was not immunosuppressive. It is suggested that to avoid cyclosporine-induced nephrotoxicity, the dosage of cyclosporine should be decreased when ketoconazole and cyclosporine are used in combination.
