Uptake of uridine and its incorporation in RNA of Sarcoma 180 ascites tumour cells following in vivo gamma radiation and mitomycin C treatment

The rate of radioactive uridine uptake and incorporation into Sarcoma 180 cell RNA was examined following the exposure of ascites tumour-bearing mice to gamma-radiation alone or in combination with mitomycin C (MMC). Gamma-irradiation alone with a dose ranging from 400 R to 1000 R resulted in an increase in the precursor pool of labeled uridine and its incorporation into TCA-insoluble product studied at varying times after the exposure exhibited a peak at 24 hr of irradiation. MMC causes a reduction in the uptake of labelled uridine as well as in the incorporation of the labeled into the bulk RNA. The most interesting feature is the observation that when MMC was administered intraperitoneally 18 hr prior to irradiation both uptake and incorporation were hampered to a larger extent. Polyacrylamide gel electrophoretic patterns revealed that synthesis of both high and low molecular weight RNA was blocked.
