Effect of indomethacin and acetylsalicylic acid (ASA) on peroxidase activity of gastric mucosa of rat

The effect of Indomethacin, Theophylline and Acetylsalicylic Acid on the histochemical activity of rat gastric mucosa has been studied. For this purpose a single dose of indomethacin (5 mg/kg), acetylsalicylic acid (25 mg/kg) or theophylline (10 mg/kg) was administered and its effects on histochemical Lactic Dehydrogenase (LD) Succinic Dehydrogenase (SD) Glucose 6-Phosphate Dehydrogenase (G-6PD) and Peroxidase reactions were evaluated and compared with their respective controls at 1,3,6 and 24 hours after treatment. The histochemical activities were evaluated by the rate of chromophore per unit of area. Our results show that both indomethacin and ASA inhibited peroxidase activity, and the maximal inhibition was observed at 6h, (49%, p < 0.02 and 38%, p < 0.05, respectively). This effect was reversed 24 h, after treatment. But theophylline, induced an increase, especially at 6 h, (38%, p < 0.05). On the other hand, indomethacin, but not acetylsalicylic acid, induced an increase (11%, p < 0.02) and a decrease (20%, p < 0.05) of LD and SD respectively. The G-6PD activity value was not modified by these drugs. Our results suggest that both prostaglandin synthesis inhibitors could induce inhibitory effects on peroxidase activity.
