Choice of DNA probe for the analysis of the polymorphism of a beta-globin cluster in thalassemia patients

Sequences containing a fragment of .beta.-globulin gene and 5'-flanking sequences, were cloned from phage .lambda.H.beta.G3 (from human gene library) carrying an insert of .beta.-globulin gene. The DNA-probes obtained were used for the analysis of length polymorphism in DNA restrictive fragments of 14 patients with .beta.-thalassemia major, and of 8 donors. No delition forms of thalassemia have been detected in the patients investigated. DNA-probe pVC-19-1,9.beta. has revealed a polymorphous site by restrictase Hinf I in three patients.
