Cyto-destruction of leukemic blasts in vitro using a monoclonal antibody and complement

The cytolytic activity of the monoclonal antibody J5 (anti-CALLA) was assessed on human radioactive chromium-labelled lymphoblasts in the presence of a twenty-fold number of mononuclear cells, with complements as effector mechanism. The lytic capability of the antibody could be assessed up to a titer of 1/40,000. A unique treatment with J5 plus complement yielded a maximum lysis of 84 per cent with antibody dilutions between 1/100 and 1/200. By means of three consecutive such treatments, 97 per cent cell lysis could be attained. The application of this method to the leukaemic cells of four different patients induced variable results ranging between 82 per cent and 100 per cent. These findings allow one to design the ideal conditions for using J5 as a means of "in vitro" elimination of residual blast cells for bone marrow transplantations.
