Correlation of acute potassium depletion, prostanoids and renal function

The relation between renal function indices and excretion of prostanoids in urine was studied in 8 healthy women during clearance periods in hypotonic polyuria (oral water load followed by 5% glucose infusion) and successive antidiureses induced with lysine-8-vasopressin (5 mU bolus followed by continuous infusion of 0.04 mU/min). Studies were made during normal potassium balance and during moderate K depletion, induced with furosemide (40 mg daily by muscle for 4 days) or a diet deficient in K, with or without indomethacin 1.73 mg/kg body weight by muscle. Correlation coefficients indicated that in K depletion there is decreased kaliuretic efficiency in diuresis and sodium excretion and inhibited Na reabsorption in diluting segments. Indomethacin treatment further decreased transport function. In all conditions prostanoid excretion and probably synthesis was independent of urinary flow. Results in normal K balance and in deficiency were consistent with the saluretic role of prostaglandin E2 in the presence of lysine-8-vasopressin. When prostanoid synthesis was inhibited by indomethacin the positive correlations between hypokaliaemia and renal antidiuresis and antisaluresis became significant.
