Study on the rapid detection of interleukin-2 biological activity with mouse thymocytes

In this paper, we describe a new method for detecting interleukin-2 (IL -2). The ConA-activated proliferation of C57BL/6 mice thymocytes is extremely sensitive to the inhibitory effect of hydrocortisone (HC), and can be completely blocked by a very low concentration of HC (10-7 mol/L). This inhibition is reversed by the addition of a 30 h PHA-activated human tonsillar lymphocyte supernatants (PHA-SN) or a 24 h ConA-activated mouse splenic cell supernatants (ConA-SN), and this abrogation of HC-blocked ConA-induced thymocyte activation by PHA-SN or ConA-SN was dose-dependent on the supernatant concentration, but not the supernatants containing the product of LPS-activated macrophages. The restoration of PHA-SN could be completely removed by absorption with human T cell blasts, and it was also found that the restoring activity of PHA-SN was greatly decreased by absorption with human B cell blasts. These results have strongly suggested that IL-2 is responsible for the restoring phenomenon. More than 20 experiments demonstrated that this method was stable and reproducible. Therefore, it is suitable for research work as well as clinical study.
