Orphenadrine plasma levels and amelioration of extrapyramidal side effects in schizophrenic patients treated with haloperidol

The aim of this study was to ascertain the possible relationship between plasma levels of an anticholinergic drug, orphenadrine, and its clinical efficacy on EPS in schizophrenic patients, diagnosed by DSM-III, treated with haloperidol. The psychopathological and extrapyramidal picture were evaluated respectively by CPRS and Simpson and Angus Rating Scale. Results show a positive relationship between EPS percentage of improvement and orphenadrine plasma levels, whereas plasma concentrations of the drug over 260 ng/ml seem to be the most effective in counteracting haloperidol induced EPS.
