Naturally or pathologically occurring brown pigmented accumulations of melanin or haemosiderins may cause problems of interpretation when diamino benzidine is used to visualise the final reaction product of immunoperoxidase procedures. A 2.5 g/l solution of potassium permanganate bleaches melanin without affecting the subsequent immuno-staining of a number of cell markers used in ophthalamic histopathology-keratin (epithelial cells), glial fibrillary acidic protein (glial cells) and factor VIII-related antigen (vascular endothelial cells). Permanganate has also been found abolish endogenous peroxidase activity.
