Hemopoietic progenitors before and after bone marrow transplantation

The value of assays for peripheral and bone marrow hemopoietic progenitors before and after bone marrow transplantation (BMT) was assessed in two patients with aplastic anemia and one with acute lymphoblastic leukemia (ALL). One allogeneic and two syngeneic BMT were performed with successful engraftment. After the bone marrow infusion, peripheral and bone marrow hemopoietic progenitors, CFU-GM, BFU-E and CFU-E, were assayed serially, using a methylcellulose culture method. In the peripheral blood, numerous hemopoietic progenitors appeared just after the bone marrow infusion, but they rapidly disappeared within three days. From Days 30 to 40, CFU-GM and BFU-E reappeared in the peripheral blood and increased to higher levels than normal, and gradually decreased to the normal range, but CFU-E were no longer observed in the peripheral blood. On the other hand, in the bone marrow, an increase of hemopoietic progenitors was observed from Days 2 to 14, and preceded the increase of conventional hematological parameters. These observations demonstrated the time course of hemopoietic reconstruction after BMT. It was suggested that the serial assay of hemopoietic progenitors is useful for early prediction of bone marrow engraftment.
