Isolation of the principal sugars of Stevia rebaudiana

The hot aqueous extract of dried leaves (80 g) of S. rebaudiana was chromatographed on Amberlite XAD-2 column using MeOH as eluent. The methanol eluate was taken to dryness to give 0.5 g of material, which was chromatographed on a column of Sephadex LH-20. This column was washed with methanol. Fractions of 20 ml were collected and checked by TLC SiO2 with 60:15:25 BuOH/AcOH/H2O. Fractions 8-9 (50 mg) contained rebaudianoside A, fractions 17-18 (60 mg) contained stevioside identified by I.R. and 1H and 13NMR spectroscopy.
