Lens protein changes in the Emory mouse cataract

Water-soluble and urea-soluble crystallins in the Emory cataractous and normal mouse lenses were investigated by using sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis with densitometric scan. At 0.5 mo. of normal lens, the 20,000 dalton component was relatively more abundant than the 28,000 dalton component. However, with increasing age at 7 mo., the relative quantities of the 28,000 dalton component increased, thus reversing the earlier proportion. The above phenomenon was observed in the water-soluble crystallin. In the urea-soluble crystallin, the 28,000 dalton component was more dominant than that of the 20,000 dalton component at 0.5 mo. of age and the 20,000 dalton component increased considerably with age at 7 mo. Further, water-soluble and urea-soluble crystallins in both the normal and the Emory mouse lenses at 7 mo. were investigated with respect to cataract development. There was no difference between the normal and the Emory mice in the water-soluble crystallin. In the crystallin from the Emory mouse cataract, the relative quantities of 28,000 and 20,000 dalton components increased slightly and the heterogeneous peaks revealed .apprx. 20,000 dalton component.
