Morphological study of dog spermatozoa

152 ejaculates of 9 Beagles were examined by 2 methods. In method A, sperm smears were stained according to the method of W. Karras (Monatshefte fur Praktische Tierheilkunde (1950) 2, 162-167) and modified by J. Kordel (Hannover, Tierarztliche Hochschule, Diss. (1959)). In method B, spermatozoa were fixed in a formol-citrate Rose Bengal solution. Using both methods, the percentages of abnormal spermatozoa were within acceptable limits. Nevertheless, some significant differences in the percentage of abnormalities were found between the 2 methods. Method B was the one recommended because acrosome structure was better differentiated, staining was more intense, and it was easier and quicker to carry out than method A.
