Effect of "764-1" eyedrops on ultrastructure of corneal scars in rabbits

"764-1" eyedrops prepared by the Institute of Hematology showed beneficial clinical effect on corneal scars. We set up a model of corneal scar in rabbits in order to study the mechanism of the "764-1" eyedrops' effects on the scar ultrastructures in rabbits and we present our findings here. These indicated that the eyedrops exerted a regulatory action on the metabolism of the scar connective tissue. The principal effect of the drug lay in its inhibition of the proliferation and function of collagen-producing cells. The collagen-producing cells in corneal scar underwent hyperplasia and hypertrophy, irregular in direction, and the collagen fibers of varying width were also in disorderly arrangement. There were amorphous substances around the cells. Fibroblastoid reticular cells and ordinary fibroblasts with developed rough endoplasmic reticulum were the two types of collagen-producing cells that proliferated in the corneal scars. After treatment the number of parenchymal cells was reduced. In appearance the fibroblasts were slender and fusiform with little cytoplasm, clear cytoplasmic membranes and an increased amount of nuclear heterochromatin. They lay parallel to the neatly aligned collagen fibers. These fibroblasts tended to grow to maturity. The collagen fibers were nearly uniform in size and the laminae formed by them ran vertical to each other. The change in distribution of the collagens effected a change in the cornea from cloudy toward translucent.
