The viability of arterially perfused enucleated rabbit eyes was demonstrated. Aqueous humor formation rate of perfused eye was similar to that of living rabbit eye. The protein concentration in the aqueous humor of perfused eye was almost the same as that in the control eye after 1 h perfusion with rabbit serum. Aqueous ascorbate level in the perfused eye was slightly higher than that in the non-perfused eye after 1 h perfusion with plasma level ascorbate solution. In this experiment system, effects of the metabolic inhibitors and ocular hypotensive drugs on aqueous humor formation were also studied. Ouabain, at the level of 10-5 M, revealed 50% inhibition in aqueous humor production. At 10-4 M, diamox lowered aqueous humor formation by 40%. Epinephrine and timolol, at a concentration of 10 .mu.g/ml reduced aqueous humor formation by 50-60%. However, pilocarpine was ineffective. An arterially perfused enucleated eye can serve as a useful model for the study of aqueous humor formation.
