Alterations in isoproterenol-induced cardiac metabolic changes by perhexiline

The effect of perhexiline pretreatment on cardiac metabolism in experimental myocardial necrosis was studied. Myocardial necrosis was produced in rats by isoprenaline 85 mg/kg i.m. for 2 days. Perhexiline was administered in doses of 3 and 10 mg/kg (i.m.) for 7 days and isoprenaline on days 6 and 7. Isoprenaline reduced the glycogen, ATP and creatine phosphate (CP) and increased the lactate level in the myocardial tissue. Serum lactate dehydrogenase (LDH) level was also raised. Perhexiline had a dose-related effect in maintaining these values near the normal range and the effect was more with a dose of 10 mg/kg. Perhexiline alone for 7 days did not alter any parameter significantly when compared with the control group. The biochemical effect of perhexiline could be correlated with the gross effect and hypertrophy of myocardium. Perhexiline may act by increasing lactate utilization and high energy phosphate pool, thus maintaining the cellular integration and preventing further damage to the myocardium.
