Correlation between in vitro chemosensitivity of human tumors and the corresponding xenografts in nude mice

Many investigators have transplanted human malignant tumors into nude mice and their studies have shown morphological and functional properties of the original tumors are retained well in the corresponding xenografts. Preservation of chemosensitivity in the xenograft is still unclear. As a means of investigating whether the chemosensitivity of the xenograft in nude mice is similar to that of the original tumor when it grows in the patient, the in vitro colony formation and chemosensitivity of patient's original tumors and the corresponding xenografts in soft agar clonogenic assay were examined. In this study morphology of the original tumors was retained well in the xenografts. The xenografts showed several times higher plating efficiencies and became more sensitive to anticancer drugs than the original tumors. The spectrum of sensitivity to anticancer drugs of original tumors was well-preserved in the corresponding xenografts. Human tumor xenografts may be used to develop quantitative chemosensitivity tests.
