Detection of auto-antibodies to zona pellucida in human sera

A total of 95 human sera was tested by indirect immuno-fluorescence technique for the possible presence of auto-antibodies to zona pellucida. The incidence of anti-zona activity in the unabsorbed sera was 71.4% in infertile, 53.3% in tubectomized, 40.0% in the non-pregnant fertile and 20.0% in the pregnant women and 26.7% in the normal fertile men. To exclude false-positive reactions due to non specific serum components, all positive sera were absorbed with procine red blood cells and the anti-zona activity was retained in 51.4% in the infertile group and 26.7% in the tubectomized women. Anti-zona activity was lost in all the positive sera of fertile group (men and women) after absorption. The positive sera of the infertile and tubectomized women were further absorbed with zona coated eggs and were retested. Fluorescence was lost in all the positive sera thus demonstrating the presence of antibodies specific to zona antigen in the infertile and tubectomized women.
