Effect of continuous cadmium administration on cytochrome P-450-dependent enzyme activities of the liver and kidney of male rats

The effect of Cd on hepatic and renal microsomal enzymes of rats was investigated by determination of cytochrome P-450 and cytochrome b5 levels, aniline hydroxylase and aminopyrine-N-demethylase activities and measurement of hexobarbital sleeping time during continuous administration by drinking water or gavage throughout 3 mo. The influence on these liver enzymes was in evidence after daily oral administration. The decrease of the cytochrome P-450 level was in correspondence with the inhibition of N-demethylase activity and prolonged hexobarbital sleeping time. Independent of the mode of administration cytochrome P-450 and cytochrome b5 levels in the kidney, decreased at a dosage of 1/10 LD50 of CdCl2. These inhibition effects were more strongly pronounced during the first 7 wk than in the following weeks of treatment. The mixed-function oxidase system was not affected at a dosage of 1/30 LD50 corresponding to a level of 50 ppm Cd in the drinking water.
