Characteristics of phosphatidylcholine/peptidylglycolipid liposomes

Physico-chemical characteristics of liposomes comprised of egg phosphatidylcholine and varying mole fractions of 2 N-acetylmuramyl dipeptides were studied. Small unilamellar liposomes were formed by sonication and fractionated by centrifugation. Inclusion of peptidylglycolipids in egg phosphatidylcholine bilayers had no effect on the size distribution and internal volume of liposomes. Unlike these observations, the leakage rates of neutral solutes from egg phosphatidylcholine/peptidylglycolipid liposomes were significantly higher than those from egg phosphatidylcholine liposomes. Peptidylglycolipids in these liposomes were preferentially localized in the outer surface. Incorporation of (peptidyl) glycolipids in phospholipid bilayers evidently perturbs the normal mode of the phospholipid packing. The transbilayer lipid distributions in sonicated vesicles comprised of binary mixtures of lipids is mainly controlled by the effective polar head group volume of the lipid.
