Characterization of the carbohydrates of Schistosoma japonicum adult worm, egg and cercaria by analysis of lectin binding and antibody reaction

The binding of the FITC-labelled lectins Con A, soybean agglutinin (SBA), wheat germ agglutinin (WGA), Dolichus biflorus agglutinin (DBA), Ulex europeaus agglutinin (UEA 1), peanut agglutinin (PNA) and Ricinus communis agglutinin (RCA 120) to cross sections of adult eggs and cercariae of S. japonicum was investigated. Most lectin binding receptors were located at the same sites as FITC-labelled antibodies reacted. Antibodies did not inhibit lectin binding, but lectins blocked the antigenic sites and weakened the antibody reactions. Con A and WGA were unselectively bound but some of the others could be used for purification of antigens: PNA for adult tegument antigens, PNA and SBA for vitelline space and vitelline membrane, RCA 120 for gut and tegument.
