Nuclear translocation of progesterone receptor--progestogen complex in vitro

Cytosol and nuclear fractions were prepared from uteri from oestrogen-primed rabbits, the cytosols were incubated in vitro with [superscript 3H]-progestogens, and the cytosol-steroid receptors so formed were combined with the nuclei, after which the receptor-steroid complexes in the nuclei were determined. The [superscript 3H]-progesterone-receptor complex in the cytosol was exchanged for cytosolic receptor sites by progesterone, dydrogesterone and norethindrone. When the steroid receptors translocated to the nucleus by such steroids in the in vitro system were determined using an exchange assay, progesterone was found to translocate the receptor to the nucleus much more frequently than did dydrogesterone or nerethindrone. It was suggested that progesterone was able to achieve the nuclear translocation of its own receptor.
