The influence of some technical factors on Ag-staining of NOR in human lymphocyte chromosomes was studied. To be completely stained, poorly stained NOR needed more time than those stained strongly. Some changes in the morphology of short arms of acrocentric chromosomes bearing NOR were detected before the NOR started to stain with Ag. The morphology of Ag-deposits in completely stained NOR was found to be specific and constant for a given NOR. A new modification of the Ag-I method of NOR staining is proposed.
