Intake of 4-methylumbelliferyl-alpha-glucoside-entrapping liposome into guinea pig macrophages in vivo

The entry of 4-methylumbelliferyl-.alpha.-glucoside-entrapping liposome into guinea pig macrophages and degradation by .alpha.-glucosidase was evaluated using fluorescence microcytophotometry. The amount of fluorescence formed by cleavage of glucosidic bond was parallel with incubation times in the peritoneal cavity. Liposomes phagocytized by macrophages apparently fuse with lysosomes and the contained 4-methylumbelliferyl-.alpha.-glucosides are broken down by .alpha.-glucosidase in lysosomes. This procedure was provided for cytochemical staining of .alpha.-glucosidase in a single macrophage.
