Rat heart mitochondria, which had respiratory control ratio of 5.5 and ADP/O ratio of 1.9 with succinate as a substrate, did not show any high-affinity bindings for Ca2+. Mitochondria satisfactory for respiratory control and ADP/O ratio were not always expected to be satisfactory for high-affinity Ca2+ binding. Different results reported on the number of high-affinity Ca2+ binding sites and properties of them are due probably to the differences in the preparations and conditions of experiments.
