Tissue distribution of guinea pig alpha 1-microglobulin

The levels of .alpha.1-microglobulin in various guinea pig organs were determined after homogenization followed by solubilization with sodium deoxycholate or sonication. Higher amounts were found in the kidney and liver than in the spleen, placenta, adrenal glands, heart, brain, lungs, abdominal musle and testes. No .alpha.1-microglobulin could be detected in solubilized lymphocytes or erythrocytes. Subcellular fractions from the kidney, liver, placenta and muscle were prepared by differential centrifugation. Liver .alpha.1-microglobulin was found in the microsomal fraction (25%) and in the 105,000 g supernatant (67.5%). Kidney, placenta and muscle .alpha.1-microglobulin were found in the 105,000 g supernatants (89, 88 and 100%, respectively). .alpha.1-Microglobulin accumulated in the medium of liver cells incubated for 4 h. The accumulation could be inhibited by 1.5 mM sodium azide.
