The effects of bilateral castration, replacement with testosterone and ascorbic acid separately and in combination, for 10 days was investigated in adult albino rats. Castration resulted in significant decrease in the androgen-sensitive parameters and histology of both proximal and distal vas deferens. The latter was comparatively more affected by castration than the proximal region. Replacement therapy with testosterone revealed that not only the weight and histology of the 2 regions, but also their biochemical constituents manifest differential sensitivity to the same dose of testosterone. Ascorbic acid administration to castrated rats simulated the effects of testosterone to some extent and combined testosterone plus ascorbate administration manifested a synergistic effect of recovery of androgenic parameters in vas deferens. Distal vas deferens possessed a higher threshold requirement of testosterone for the maintenance of its structural and biochemical integrity than the proximal vas deferens.
