Microtechnique for leukocyte culture from whole blood without serum supplementation

Under 7 possible experimental groups 350 blood cultures were put out of 50 blood samples from same number of healthy donors, using less rich and rich medium (MEM ) without any serum supplementation. The growth potential was assessed using mitotic index as a parameter. The .chi.2 comparisons between each group in various permutation and combinations and their P values were worked out. The individual mitotic index values within a group did not show significant differences. The present simple and cheap procedure developed for the whole blood culture, foregoing serum supplementation, required no special handling and proved that the defined media by itself without the addition of serum proteins can support the growth of such cultures. The non-serum-supplemented blood cultures, termed as serum free whole-blood culture, procedure is towards an approximation to the in vivo condition.
